Tigecycline therapy is associated with a significant risk of INR prolongation, affecting over a third of patients. Regular monitoring of International Normalized Ratio (INR) is recommended during tigecycline treatment.
Area of Science:
- Pharmacology
- Clinical Medicine
- Infectious Diseases
Background:
- Tigecycline is a broad-spectrum antibiotic used for complicated infections.
- Potential drug-drug interactions and adverse effects require careful consideration.
- International Normalized Ratio (INR) monitoring is crucial for patients on anticoagulant therapy.
Purpose of the Study:
- To determine the incidence of International Normalized Ratio (INR) abnormalities in patients receiving tigecycline.
- To assess the impact of tigecycline on INR values in a hospitalized patient cohort.
Main Methods:
- Retrospective review of 79 patients treated with tigecycline between June and September 2016.
- Comparison of INR values before and after tigecycline therapy.
- Analysis of INR abnormality rates in monotherapy versus combination therapy.
Main Results:
- Out of 60 evaluable patients, 19 (34.5%) experienced INR prolongation during tigecycline treatment.
- INR prolongation was mild in 12, moderate in six, and severe in one patient.
- Discontinuation of tigecycline led to INR normalization in 10 patients; no significant difference in INR abnormality rates between monotherapy and combination therapy.
Conclusions:
- Tigecycline therapy is frequently associated with INR prolongation (34.6%).
- Regular INR monitoring is advisable for patients undergoing tigecycline treatment.
- Further studies may be needed to elucidate the mechanisms of tigecycline-induced INR changes.
