The platelet phase, the second stage of hemostasis, commences around 15-20 seconds after an injury. It follows and overlaps with the vascular phase, during which blood vessels constrict to minimize blood loss.

Summary

Wheat germ agglutinin (WGA) and Phaseolus vulgaris agglutinin (PHA) induce platelet oxidative stress and impair energy metabolism. Lens culinarian agglutinin (LCA) is ineffective, while WGA is more potent than PHA.

Background:

Purpose of the Study:

  • To investigate the impact of lectins (WGA, PHA, LCA) on platelet oxidative stress.
  • To determine the effects of these lectins on platelet aerobic metabolism and energy production.

Main Methods:

  • Measurement of reactive oxygen species (ROS), superoxide anion, and lipid peroxidation.
  • Assessment of aerobic metabolism, including oxygen consumption, ATP synthesis, and P/O values.
  • Evaluation of platelet membrane potential and proton gradient formation.

Main Results:

  • WGA and PHA significantly increased ROS, superoxide anion, and lipid peroxidation in platelets; LCA had no effect.
  • WGA demonstrated greater potency than PHA in inducing oxidative stress.
  • WGA and PHA acted as uncoupling agents, increasing oxygen consumption and decreasing ATP synthesis, impairing the proton gradient.
  • NADPH oxidase 1, syk, and PI3K were implicated in lectin-induced oxidative stress.

Conclusions:

  • Lectins, particularly WGA, induce significant oxidative stress in platelets.
  • These lectins disrupt platelet energy availability by impairing aerobic metabolism and membrane integrity.
  • The observed metabolic dysfunction and oxidative stress may lead to platelet death, indicated by decreased metabolic activity and increased LDH release.
