Background:

  • Traumatic shock is a critical condition with complex pathophysiological mechanisms.
  • Platelet-activating factor (PAF) is a potent lipid mediator implicated in inflammatory and immune responses.

Purpose of the Study:

  • To investigate the formation of platelet-activating factor (PAF) in peritoneal fluid following traumatic shock in rats.
  • To determine the functional role of PAF in the pathogenesis of traumatic shock.

Main Methods:

  • Induction of traumatic shock in rats using Noble-Collip drum trauma.
  • Analysis of peritoneal fluid using high-performance liquid chromatography (HPLC).
  • Assessment of platelet aggregation in response to peritoneal fluid extracts and blockade with a specific PAF receptor antagonist (CV-6209).

Main Results:

  • Peritoneal fluid from traumatized rats contained a phospholipid with HPLC retention time similar to authentic PAF, absent in control rats.
  • Peritoneal fluid extracts from traumatized rats induced significant aggregation of washed rabbit platelets.
  • PAF receptor antagonist CV-6209 blocked the platelet aggregation induced by these extracts.
  • Platelet aggregation activity in peritoneal fluid increased nearly four-fold in traumatized rats compared to sham-shock controls.

Conclusions:

  • Findings are consistent with the formation of platelet-activating factor (PAF) during traumatic shock in rats.
  • The data support a significant role for PAF in the pathogenesis of traumatic shock, evidenced by its formation and platelet-aggregating activity.
