Summary

Mesenteric lymph node nuocytes promote allergic rhinitis in mice. These cells, stimulated by IL-25 and IL-33, produce key cytokines, driving allergic responses and eosinophilia.

Area of Science:

  • Immunology
  • Type 2 Immunity
  • Allergic Inflammation

Background:

  • Nuocytes are crucial for Type 2 immunity.
  • The specific role of innate lymphoid cells group 2 (ILC2s) in allergic rhinitis is not fully understood.

Purpose of the Study:

  • To investigate the function of nuocytes from mesenteric lymph nodes in allergic responses.
  • To determine the impact of nuocytes on murine models of allergic rhinitis.

Main Methods:

  • Nuocytes were isolated from wild-type and double-deficient mice (Il17br-/-Il1rl1-/-) after IL-25 and IL-33 stimulation.
  • Cytokine production (IL-5, IL-13) by nuocytes was assessed.
  • Mesenteric lymph node-derived nuocytes were adoptively transferred into a mouse model of allergic rhinitis.

Main Results:

  • Stimulated wild-type nuocytes produced IL-5 and IL-13.
  • Adoptive transfer of wild-type nuocytes exacerbated allergic rhinitis symptoms, including sneezing, nasal rubbing, and eosinophilia.
  • Nuocytes from double-deficient mice did not induce these allergic responses.

Conclusions:

  • Nuocytes from mesenteric lymph nodes play a significant role in promoting allergic rhinitis.
  • The IL-17BR and IL1RL1 pathways are critical for nuocyte-mediated allergic responses.
