Area of Science:

  • Endocrinology
  • Pharmacology

Background:

  • Glucocorticoids have profound effects on leukocyte counts.
  • RU 486 is known to possess antiglucocorticoid properties.

Purpose of the Study:

  • To evaluate the peripheral antiglucocorticoid activity of RU 486 in humans.
  • To determine if RU 486 can antagonize acute glucocorticoid effects on blood leukocyte counts.

Main Methods:

  • A double-blind, cross-over, latin-square design study was conducted on eight normal male subjects.
  • Subjects received oral RU 486 (or placebo) and dexamethasone (or placebo) at specified times.
  • Blood leukocyte counts (eosinophils, lymphocytes, neutrophils) were measured before and after treatments.

Main Results:

Related Experiment Videos

  • Dexamethasone significantly altered leukocyte counts (eosinophil and lymphocyte decrease, neutrophil increase).
  • RU 486, alone or in combination with dexamethasone, did not significantly alter leukocyte counts compared to placebo.
  • RU 486 administration inhibited the dexamethasone-induced changes in leukocyte counts.
  • Conclusions:

    • RU 486 exhibits peripheral antiglucocorticoid activity in humans by inhibiting dexamethasone-induced leukocyte count alterations.
    • This simple leukocyte count assay is a valuable tool for assessing peripheral antiglucocorticoid activity.
