Summary

This study determined cefixime bioavailability in healthy volunteers. Oral cefixime absorption was similar for 200-mg and 400-mg doses, with absolute bioavailability ranging from 40-52%.

Area of Science:

  • Pharmacokinetics
  • Drug Absorption and Metabolism

Background:

  • Understanding the absolute bioavailability of cefixime is crucial for optimizing oral dosage regimens.
  • Cefixime is a third-generation cephalosporin antibiotic used to treat various bacterial infections.

Purpose of the Study:

  • To determine the absolute bioavailability of cefixime following oral administration in healthy volunteers.
  • To compare the absorption of cefixime from oral solutions and capsules at different dosages.

Main Methods:

  • A four-way crossover study involving 16 healthy volunteers.
  • Administration of single 200-mg intravenous (IV) and oral solution doses, plus 200-mg and 400-mg capsule doses.
  • Collection of blood and urine samples for 24 hours to measure cefixime concentrations.

Related Experiment Videos

Main Results:

  • Cefixime was well-tolerated with no serious adverse effects observed.
  • Absolute bioavailability based on serum AUC ranged from 40.2% to 52.3% for oral doses.
  • Elimination half-life was consistent across all administration routes, approximately 3.2-3.5 hours.

Conclusions:

  • The percentage of cefixime adsorbed after 200-mg and 400-mg oral doses was found to be similar.
  • These findings support the use of oral cefixime formulations with predictable absorption profiles.
