Aqueous humor causes ocular lymphatic vessels to regress. This finding, observed in real-time live imaging, may lead to new treatments for lymphatic diseases.
Area of Science:
- Ophthalmology
- Vascular Biology
- Cell Biology
Background:
- Lymphatic vessels are crucial for fluid homeostasis and immune surveillance in the eye.
- Dysfunctional ocular lymphatic vessels are implicated in various eye diseases.
Purpose of the Study:
- To investigate the effect of aqueous humor on pre-existing conjunctival lymphatic vessels.
- To determine if aqueous humor can induce regression of ocular lymphatic vessels in vivo and in vitro.
Main Methods:
- Aqueous humor from bovine or murine eyes was injected into Prox-1-GFP transgenic mice.
- Live in vivo imaging was used to observe conjunctival lymphatics.
- Ex vivo tissue cultures and human microdermal lymphatic endothelial cell cultures were performed to assess lymphatic tube regression.
Main Results:
- Aqueous humor induced regression of lymphatic vessels in normal conjunctiva and inflamed corneas.
- In vitro studies confirmed that aqueous humor causes regression of lymphatic endothelial cell tubes.
Conclusions:
- This study provides the first direct live imaging evidence that aqueous humor induces lymphatic regression.
- These findings suggest potential new therapeutic strategies for ocular and systemic lymphatic diseases.
