Background:

Purpose of the Study:

  • To investigate the transcriptional profile of chicken embryonic fibroblasts (DF1) stimulated by chicken MDA5 (chMDA5) and poly(I:C).
  • To identify interferon-stimulated genes (ISGs) upregulated by chMDA5 activation.
  • To evaluate the antiviral potential of identified ISGs against avian influenza virus H5N6.

Main Methods:

  • RNA sequencing (RNA-seq) to compare transcriptional profiles of DF1 cells transfected with chMDA5 and poly(I:C) against controls.
  • Real-time PCR to validate the upregulation of nine key ISGs.

  • These findings offer insights for developing novel chicken antiviral therapies targeting MDA5-mediated immunity.
