Nitric oxide synthase (NOS) isoforms are present in ram sperm and change during capacitation. Melatonin affects NOS localization but not nitric oxide (NO·) levels, suggesting NO· metabolism isn't melatonin's primary role in ram sperm capacitation.
Area of Science:
- Reproductive Biology
- Sperm Physiology
- Molecular Endocrinology
Background:
- Nitric oxide (NO·), synthesized by nitric oxide synthase (NOS), plays a role in sperm function.
- NOS activity increases during sperm capacitation, a process essential for fertilization.
Purpose of the Study:
- To determine the presence, localization, and changes of NOS isoforms in ram spermatozoa during in vitro capacitation.
- To investigate the effect of melatonin on NOS expression, localization, and NO· levels in capacitated ram spermatozoa.
Main Methods:
- Western blot analysis to detect NOS protein expression.
- Indirect immunofluorescence (IFI) to visualize NOS isoform localization.
- DAF-2-DA/PI staining to quantify NO· levels.
- In vitro capacitation of ram spermatozoa with and without melatonin.
Main Results:
- Neuronal NOS (nNOS) and epithelial NOS (eNOS) proteins were detected by Western blot; all three isoforms (nNOS, eNOS, inducible NOS/iNOS) were detected by IFI, with varying immunotypes during capacitation.
- NO· levels increased post-capacitation, and L-arginine supplementation enhanced NO· production and acrosome reaction.
- Melatonin altered NOS distribution (IFI) but not protein levels (Western blot) and did not affect NO· levels.
Conclusions:
- NOS isoforms are present and dynamically regulated in ram spermatozoa during capacitation.
- Melatonin's effect on ram sperm capacitation is unlikely to be mediated through NO· metabolism.
