Summary

Canine osteosarcoma (OS) exhibits higher miR-93-5p expression than human OS. Inhibiting miR-93-5p significantly impacted canine OS cell proliferation and apoptosis more than human OS cells.

Area of Science:

  • Comparative oncology
  • Molecular biology
  • Biomedical research

Background:

  • Osteosarcoma (OS) is a common bone cancer in dogs and humans with shared characteristics.
  • Understanding biological differences in OS progression can improve patient outcomes.

Purpose of the Study:

  • To compare microRNA (miRNA) expression in human and canine OS.
  • To investigate the role of the miR-106b-25 cluster in OS progression.
  • To develop a comparative model for OS treatment.

Main Methods:

  • Compared expression of miR-106b, miR-25, and miR-93-5p in human and canine OS tissues.
  • Evaluated the functional response of human (143B) and canine (DAN) OS cells to miRNA inhibition.

Main Results:

  • miR-93-5p expression was elevated in all OS specimens, with higher levels in canine OS.
  • Canine OS cells showed a more significant response to miR-93-5p inhibition, affecting proliferation and apoptosis.
  • p21 expression increased post-inhibition, with weaker staining in canine OS.

Conclusions:

  • Canine OS demonstrates higher miR-93-5p levels compared to human OS.
  • miR-93-5p inhibition had a more pronounced functional effect on canine OS cells.
  • This comparative study highlights potential therapeutic targets for osteosarcoma.
