Background:

  • Phosphate toxicity is a significant cardiovascular risk in chronic kidney disease (CKD).

Purpose of the Study:

  • To develop a miniaturized, rapid calcification propensity test for phosphate toxicity assessment.
  • To improve the T50 test for faster, bedside or in-line measurements.

Main Methods:

  • A microfluidic chip was developed using ultrasonic hot embossing.
  • The chip integrated a laser diode, light-dependent resistor, and Peltier element for temperature control.
  • Assay temperature was increased to 75°C, and micro mirrors enhanced signal detection.

Main Results:

  • The T50 test time was reduced 36-fold, from 381 minutes to 10.5 minutes.
  • The signal-to-noise ratio improved 9-fold due to micro mirrors.
  • The chip-based assay demonstrated speed and reproducibility.
