    Summary

    Gram-negative sepsis endotoxin directly interacts with human platelets, causing membrane changes and releasing serotonin. This interaction may explain vascular and hemostatic disorders in sepsis.

    Background:

    • Thrombocytopenia is a common complication of Gram-negative sepsis.
    • Animal studies suggest endotoxin targets blood platelets.

    Purpose of the Study:

    • To investigate the effect of Salmonella enteritidis lipopolysaccharide B (endotoxin) on human platelets.
    • To elucidate the mechanism of endotoxin-platelet interaction in sepsis.

    Main Methods:

    • Human platelets were isolated using Sepharose 2B filtration and albumin gradient.
    • Platelet response to endotoxin was assessed by measuring [3H]serotonin and adenine nucleotide release.
