Schizophrenia patients not on antipsychotics show reduced alpha5-GABA-A receptor availability in the hippocampus, suggesting GABA signaling deficits contribute to the disorder. This reduction correlates with symptom severity in these patients.

Conclusions:
- Antipsychotic-free schizophrenia patients exhibit reduced hippocampal α5-GABAARs availability in vivo.
- These findings support the hypothesis that GABA hypofunction is implicated in schizophrenia pathophysiology.
- The results highlight the potential role of α5-GABAARs in schizophrenia symptoms and disease mechanisms.
