Summary

Angiotensin II infusion causes hypertension and vascular damage. This study reveals vascular aging, involving insulin-like growth factor 1 and adiponectin, as a key mechanism in this process.

Area of Science:

  • Cardiovascular Biology
  • Molecular Medicine
  • Hypertension Research

Background:

  • Angiotensin II (Ang II)-induced vascular damage is a significant risk factor for hypertension.
  • The precise molecular mechanisms underlying Ang II-induced vascular damage remain incompletely understood.

Purpose of the Study:

  • To investigate the novel molecular mechanisms involved in Angiotensin II-induced hypertension.
  • To explore the role of vascular aging in the development of Ang II-mediated hypertension.

Main Methods:

  • Transcriptome sequencing of mouse aortic media following 28-day Ang II infusion.
  • Differential gene expression analysis, Gene Ontology (GO), and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis.
  • Validation of key differentially expressed genes using quantitative real-time PCR.

Main Results:

  • A total of 773 differentially expressed genes were identified in the Ang II-induced hypertension mouse model.
  • Functional analysis highlighted pathways related to extracellular matrix, inflammation, immune response, and notably, vascular aging.
  • Significantly increased expression levels of insulin-like growth factor 1 and adiponectin were observed, correlating with vascular aging.

Conclusions:

  • Vascular aging is identified as a critical component in Angiotensin II-induced hypertension.
  • Insulin-like growth factor 1 and adiponectin emerge as potential key candidate genes contributing to Ang II-mediated vascular aging and hypertension.
