Summary

Hyaluronic acid receptors CD44 and RHAMM show a correlation in the developing human lung at birth. This relationship is linked to lung maturation, particularly during the late saccular stage, but postnatal factors may influence expression in preterm infants.

Background:

  • Hyaluronan (HA) receptors CD44 and RHAMM (CD168) are crucial for cellular processes like proliferation, differentiation, and motility.
  • Previous research indicates a correlation between HA and RHAMM expression in neonatal lungs with gestational age (GA) and lung air content.

Purpose of the Study:

  • To investigate the relationship between CD44 and RHAMM expression in the developing human lung.
  • To determine how this relationship correlates with gestational age, postnatal age, and lung air content.

Main Methods:

  • Analysis of CD44 immunofluorescence in postmortem lung samples from 93 infants (22-41 GA) using digital image analysis.
  • Hierarchical clustering of CD44 expression with RHAMM expression, lung air, and HA content.

Main Results:

  • Five infant groups were defined based on RHAMM/CD44 expression, GA, and postnatal age (PNA).
  • A linear correlation between CD44 and RHAMM was observed in moderately preterm to term infants (31-41 GA).
  • In extremely to very preterm infants (22-31 GA), high CD44 and low RHAMM correlated with higher PNA and lung air content, independent of HA and GA.

Conclusions:

  • A linear correlation between CD44 and RHAMM expression is present during the late saccular phase of lung development at birth.
  • Postnatal influences on CD44 and RHAMM expression cannot be excluded in extremely to very preterm infants.
