Long noncoding RNA H19 (lncRNA H19) promotes arterial calcification by driving vascular smooth muscle cell transition. Inhibiting lncRNA H19 may offer a new diagnostic and therapeutic strategy for arterial calcification.
Area of Science:
- Cardiovascular Biology
- Molecular Biology
- Epigenetics
Background:
- Arterial calcification (AC) is a complex process often driven by the osteoblastic transition of vascular smooth muscle cells (VSMCs).
- Long noncoding RNAs (lncRNAs) are emerging as critical regulators in various biological processes, including cellular differentiation and disease.
- The role of lncRNA H19 in VSMC transition and AC remains incompletely understood.
Purpose of the Study:
- To investigate the role of lncRNA H19 in VSMC phenotypic transition.
- To elucidate the molecular mechanisms by which lncRNA H19 influences AC.
- To explore the potential of lncRNA H19 as a biomarker or therapeutic target for AC.
Main Methods:
- VSMCs were induced to calcify using β-glycerophosphate.
- Quantitative PCR (qPCR) and Western Blot were employed to assess gene and protein expression levels.
- Small interfering RNA (siRNA) was used to knockdown lncRNA H19 expression.
- Signaling pathway analysis focused on MAPK pathways (p38 and ERK1/2).
Main Results:
- lncRNA H19 expression was significantly upregulated in calcified VSMCs compared to normal VSMCs.
- Knockdown of lncRNA H19 reduced VSMC calcification and the expression of key osteogenic markers like Runx2 and OSX.
- lncRNA H19 was found to promote VSMC calcification through the activation of p38 MAPK and ERK1/2 signaling pathways.
Conclusions:
- The lncRNA H19/Runx2 axis plays a crucial role in promoting VSMC transition and arterial calcification via the MAPK pathway.
- lncRNA H19 represents a novel regulator in the context of AC.
- These findings suggest lncRNA H19 could serve as a potential early diagnostic marker and therapeutic target for arterial calcification.
