Pathogenic variants in the polymerase gamma gene (POLG) cause POLG-related disorders. Brain imaging in children revealed common thalamic and perirolandic signal changes, often progressing rapidly, suggesting these findings may be early biomarkers.

- POLG-related disorders in children are characterized by progressive brain abnormalities, particularly affecting the thalamus and occipital lobes.
