    Summary

    Human tonsil lymphocytes can mount an immune response against sheep red blood cells (SRBC). Adding allogeneic tonsil cells or supernatant significantly enhanced this response, even when lymphocytes alone failed to react.

    Background:

    • Investigating the immune response capabilities of human tonsil lymphocytes.

    Purpose of the Study:

    • To assess the capacity of human tonsil lymphocytes to generate an anti-sheep red blood cell (anti-SRBC) response.
    • To determine the effect of allogeneic cells, supernatant, and conditioned medium on this immune response.

    Main Methods:

    • Culture of human tonsil lymphocytes in conventional and microcultures.
    • Assessment of plaque-forming cells (PFC) as a measure of anti-SRBC response.
    • Addition of allogeneic tonsil cells (irradiated or intact), supernatant, or conditioned medium.

    Main Results:

    • Approximately two-thirds of tonsils showed a significant anti-SRBC PFC response.
    • Addition of allogeneic tonsil cells or supernatant markedly improved the response, enabling previously unresponsive lymphocytes to react.
    • An anti-SRBC response was observed even without SRBC in the culture, suggesting potential autoreactivity or cross-reactivity.

    Conclusions:

    • Human tonsil lymphocytes possess the inherent ability to mount an anti-SRBC response.
    • Allogeneic factors, such as cells and supernatant, play a crucial role in augmenting lymphocyte activation and antibody production.
    • The frequency of anti-SRBC specific B cells in tonsil lymphocytes is estimated to be 1 in 60,000.
