Super-resolution Fluorescence Microscopy01:37

Super-resolution Fluorescence Microscopy

12.1K
Super-resolution fluorescence microscopy (SRFM) provides a better resolution than conventional fluorescence microscopy by reducing the point spread function (PSF). PSF is the light intensity distribution from a point that causes it to appear blurred. Due to PSF, each fluorescing point appears bigger than its actual size, and it is the PSF interference of nearby fluorophores that causes the blurred image. Various approaches to achieving higher resolution through SRFM have recently been...

Total Internal Reflection Fluorescence Microscopy01:05

Total Internal Reflection Fluorescence Microscopy

10.9K
Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. TIRF only allows fluorophores in proximity to the solid surface to be excited. When light from a medium with a lower refractive index (such as air) hits the glass coverslip at a critical angle, the light undergoes total internal reflection stead of passing through the glass.

Area of Science:

  • Biomedical optics
  • Medical imaging technologies
  • Fluorescence microscopy

Background:

  • Near-Infrared II (NIR-II) fluorescence imaging offers deep tissue visualization due to reduced scattering and autofluorescence.
  • Existing NIR-II methods struggle to balance field of view, resolution, penetration depth, and optical sectioning.

Purpose of the Study:

  • To develop a novel NIR-II fluorescence mesoscopy system overcoming current limitations.
  • To enable high-performance deep tissue imaging with enhanced capabilities.

Main Methods:

  • Implementation of an f-θ scanning scheme for mesoscopic imaging.
  • Integration of confocal detection for optical sectioning and high resolution.
  • Utilizing NIR-II fluorescent dyes for imaging.

Main Results:

  • The system images samples up to 7.5×7.5 mm² with a lateral resolution of 6.3 µm.
  • Achieved depth-resolved imaging with zooming capabilities.
  • Successfully demonstrated in vivo cerebrovascular imaging in a mouse model of local ischemia.

Conclusions:

  • The developed NIR-II mesoscopy system provides a powerful tool for deep tissue imaging.
  • Offers new possibilities for studying cerebrovascular diseases and their mechanisms.
