Area of Science:

  • Reproductive biology
  • Cryobiology
  • Spermatozoa preservation

Background:

  • Slow freezing is standard for sperm cryopreservation but negatively impacts sperm function and structure.
  • Vitrification offers better cryoprotection for embryos, but its effectiveness for spermatozoa is limited by low recovery rates.

Purpose of the Study:

  • To develop and evaluate a modified vitrification method for native spermatozoa.
  • To compare the efficacy of this modified vitrification technique against slow freezing for sperm preservation.

Main Methods:

  • A modified vitrification protocol was developed for native spermatozoa.
  • Semen samples were divided into fresh, slow freezing, and vitrification groups.
  • Sperm vitality, motility, morphology, DNA integrity, and acrosome reaction were assessed.

Main Results:

  • Vitrification demonstrated significantly higher recovery rates, motility, morphology, and curve line velocity compared to slow freezing.
  • DNA fragmentation was reduced, and acrosome protection was improved with vitrification.
  • Principal component analysis indicated vitrification preserves sperm parameters closer to fresh samples.

Conclusions:

  • The modified vitrification method offers improved cryopreservation of spermatozoa compared to slow freezing.
  • Vitrification achieved a 65.8% recovery rate with enhanced preservation of key sperm quality parameters.
  • While both methods impact sperm, vitrification presents a more effective approach for maintaining sperm integrity.
