Background:

  • Global antibiotic resistance necessitates simple, cost-effective susceptibility testing.
  • Matrix-assisted laser desorption/ionization time of flight mass spectrometry (VITEK MS) is standard for bacterial identification.

Purpose of the Study:

  • To evaluate two novel assays for rapid and accurate bacterial antibiotic susceptibility detection using VITEK MS.
  • To assess the reliability of these assays in differentiating susceptible and non-susceptible bacterial strains.

Main Methods:

  • Development of a liquid extraction method and a direct-on-target microdroplet growth assay (DOT-MGA).
  • Testing of susceptible and non-susceptible strains of Staphylococcus aureus, Enterococcus species, Escherichia coli, and Klebsiella pneumoniae.

Main Results:

  • The liquid extraction method and DOT-MGA reliably differentiated between susceptible and non-susceptible bacterial strains.
  • Consistent results were obtained across tested bacterial species.

Conclusions:

  • VITEK MS combined with the developed assays provides a rapid and accurate method to supplement traditional susceptibility testing.
  • Clinical implementation could decrease patient care costs and expedite critical treatment delivery.
