Alternative RNA Splicing02:18

Alternative RNA Splicing

24.4K
Alternative RNA splicing is the regulated splicing of exons and introns to produce different mature mRNAs from a single pre-mRNA. Unlike in constitutive splicing where a single gene produces a single type of mRNA, alternative splicing allows an organism to produce multiple proteins from a single gene and plays an important role in protein diversity.
There are five types of alternative RNA splicing that vary in the ways the pre-mRNA segments are removed or retained in the mature mRNA. The first...

Area of Science:

  • Neuroscience
  • Molecular Biology
  • Genomics

Background:

  • Alternative polyadenylation (APA) influences mRNA fate but its neuronal heterogeneity at single-nucleus resolution is understudied.
  • APA is regulated during development and neuronal activation, yet its specific variations and regulatory mechanisms in neuronal populations remain unclear.

Purpose of the Study:

  • To investigate dynamic APA usage in distinct human neuronal cell types using single-nucleus resolution.
  • To identify regulatory mechanisms and functional implications of APA variation in neuronal populations.

Main Methods:

  • Analysis of single-cell 3' RNA sequencing data from 1424 human brain cells.
  • Motif analysis and gene functional enrichment analysis.

Main Results:

  • Distal APA sites are globally favored in neuronal cells and show usage variation between excitatory and inhibitory neurons.
  • Genes with neuron-enhanced distal polyadenylation site (PAS) usage are enriched for RNA-binding protein (RBP) binding sites and neuronal functions.

Conclusions:

  • APA regulation is extensively involved in neuronal populations at the single-nucleus level.
  • Findings provide novel insights into the roles of APA in specific neuronal cell types and potential utility for future functional studies.
