Summary

Prostaglandin E (PGE) may not be as immunosuppressive in animal models as previously thought. This study found a PGE derivative improved survival in some models and did not impair cell-mediated immunity in others.

Background:

  • Prostaglandin E (PGE) is implicated in immunosuppression after trauma and burns.
  • In vitro studies show PGE impairs leukocyte functions.

Purpose of the Study:

  • To investigate the in vivo effects of a novel long-acting PGE derivative (16,16-dimethyl-PGE) on immune function.
  • To determine if PGE's immunosuppressive effects observed in vitro translate to in vivo models.

Main Methods:

  • Administration of 16,16-dimethyl-PGE in various animal models including burn sepsis and Escherichia coli peritonitis.
  • Assessment of mortality, survival times, neutrophil migration, and cell-mediated immunity (including lymphocyte function).

Main Results:

  • The PGE derivative did not affect mortality in burn sepsis models.

  • Neutrophil migration was impaired at lower dosages, but cell-mediated immunity was not impaired and lymphocyte function improved at certain dosages in a rat burn model.
