Area of Science:

  • Food Science
  • Analytical Chemistry
  • Animal Science

Background:

  • Accurate quantification of intramuscular fat (IMF) is crucial for assessing beef nutritional quality.
  • Traditional methods like the Folch et al. (1957) assay require significant sample amounts, solvent use, and time.
  • Developing faster, more efficient analytical methods for IMF is essential for research and industry.

Purpose of the Study:

  • To evaluate two alternative methods for intramuscular fat quantification: a miniaturized Folch's method and a near-infrared (NIR) spectroscopic method.
  • To compare the performance and acceptability limits of these alternative methods against the reference Folch method using accuracy profiles.
  • To define the validity domains and calculate bias correction equations for the alternative methods.

Main Methods:

  • Miniaturized Folch's method and near-infrared (NIR) spectroscopy were employed for intramuscular fat analysis.
  • Accuracy profiles were utilized to assess method performance and establish acceptability limits.
  • Bias correction equations were derived, and measurement uncertainties were determined.

Main Results:

  • The miniaturized Folch's method demonstrated a validity domain of 1.9–13.8 g total lipids/100 g tissue.
  • The NIR spectroscopic method showed a validity domain of 4.8–13.8 g total lipids/100 g tissue.
  • Both methods exhibited less than 20% difference from the reference method and confirmed continuity with established results.

Conclusions:

  • The miniaturized Folch's method is suitable for detailed intramuscular fat quantification.
  • The NIR spectroscopic method is effective for rapid classification of large sample sets.
  • Both validated methods offer efficient, reliable alternatives to the traditional Folch assay for beef IMF analysis.
