Summary

Interleukin-1 causes joint inflammation and proteoglycan depletion in rabbits. This study investigated if immune cells cause this cartilage damage, finding that interleukin-1 still induced damage even without these cells.

Background:

  • Interleukin-1 (IL-1) is a key inflammatory cytokine involved in joint inflammation.
  • IL-1 injection into rabbit knees causes transient synovitis and cartilage proteoglycan depletion.
  • The role of infiltrating leukocytes in IL-1-induced cartilage damage remains unclear.

Purpose of the Study:

  • To determine if cartilage proteoglycan depletion induced by IL-1 is dependent on infiltrating leukocytes.
  • To investigate the effects of intra-articular IL-1 in rabbits with depleted circulating leukocytes.

Main Methods:

  • Rabbits were depleted of circulating polymorphonuclear leukocytes (PMNs) and monocytes using nitrogen mustard.
  • Interleukin-1 was injected into the knee joints of these depleted rabbits.
  • Cartilage proteoglycan content and synovitis were assessed.

Main Results:

  • Interleukin-1 injection induced synovitis and proteoglycan depletion even in rabbits depleted of circulating leukocytes.
  • The depletion of cartilage proteoglycans was not solely dependent on the presence of infiltrating PMNs and monocytes.

Conclusions:

  • Interleukin-1 can directly induce cartilage matrix degradation independent of infiltrating leukocytes.
  • These findings suggest direct effects of IL-1 on chondrocytes or the cartilage matrix contribute to joint damage in synovitis.
