The tumor suppressor p14ARF (alternative reading frame) helps maintain chromosome stability. Restoring p14ARF in cancer cells prevented the generation and proliferation of aneuploid cells, supporting its tumor-suppressive function.
Area of Science:
- Oncology
- Molecular Biology
- Genetics
Background:
- The CDKN2a locus encodes p14ARF, a tumor suppressor frequently inactivated in human cancers.
- p14ARF inhibits oncogene-induced cell cycle progression and apoptosis, primarily through p53.
- Emerging evidence suggests a role for p14ARF in maintaining chromosome stability.
Purpose of the Study:
- To investigate the role of p14ARF in chromosome stability.
- To determine if p14ARF counteracts aneuploidy, a hallmark of cancer.
Main Methods:
- Generated a monoclonal HCT116 cell line stably expressing p14ARF cDNA using a piggyback vector.
- Induced aneuploidy in HCT116 cells by treatment with the CENP-E inhibitor GSK923295.
- Assessed the effect of p14ARF re-expression on cellular ploidy.
Main Results:
- Ectopic re-expression of p14ARF in HCT116 cells significantly reduced aneuploidy.
- Cells with restored p14ARF expression exhibited a near-diploid phenotype after aneuploidy induction.
- These findings confirm p14ARF's ability to counteract the generation and proliferation of aneuploid cells.
Conclusions:
- p14ARF plays a crucial role in maintaining chromosome stability.
- The chromosome-stabilizing function of p14ARF contributes to its tumor-suppressive activity.
- Targeting p14ARF pathways could offer new therapeutic strategies for cancer treatment.
