Area of Science:

  • Reproductive biology
  • Cellular reprogramming
  • Endocrinology

Background:

  • Fibroblasts can be converted to Leydig cells via transcription factors.
  • Transgenic methods raise safety concerns for clinical applications.

Purpose of the Study:

  • To develop a safe and effective method for converting fibroblasts into Leydig-like cells.
  • To evaluate the functionality and therapeutic potential of chemically induced Leydig-like cells.

Main Methods:

  • Fibroblasts were exposed to a combination of forskolin, 20α-hydroxycholesterol, luteinizing hormone, and SB431542.
  • Gene expression profiles and testosterone production were analyzed.
  • Reprogramming efficiency in human periodontal ligament fibroblasts was assessed.

Main Results:

  • Chemical compound-induced Leydig-like cells (CiLCs) were generated.
  • CiLCs expressed key steroidogenic genes and showed gene expression profiles similar to progenitor Leydig cells.
  • CiLCs survived in testes and produced testosterone rhythmically, applicable to human cells.

Conclusions:

  • Direct reprogramming of fibroblasts into Leydig-like cells is achievable using chemical compounds.
  • This chemical induction strategy bypasses transgenic limitations, offering a safe approach for Leydig cell therapy.
  • The method preserves hypothalamic-pituitary-gonadal axis function.
