Background:

  • Choroideremia (CHM) is a rare genetic disorder causing progressive vision loss.
  • Accurate quantification of retinal pigment epithelium (RPE) area is crucial for monitoring CHM progression and treatment efficacy.
  • Existing methods for RPE area measurement may lack sufficient reliability and efficiency.

Purpose of the Study:

  • To develop and validate a reliable and efficient Photoshop-based method for quantifying preserved RPE area in CHM patients.
  • To compare the performance of the new method against the established Heidelberg Eye Explorer 2 (HEYEX2) software.
  • To assess the test-retest reliability of the Photoshop-based RPE quantification.

Main Methods:

  • Fundus autofluorescence images from CHM patients were acquired at baseline and 1-year follow-up.
  • A novel Photoshop-based image analysis technique was developed for RPE area measurement.

  • This accurate quantification tool is vital for evaluating therapeutic efficacy in CHM clinical trials.
