This study developed simvastatin-loaded nanoemulsions (SIM-LN) with excellent encapsulation and stability. Preliminary tests showed SIM-LN are non-toxic, indicating their potential for therapeutic applications.
Area of Science:
- Pharmaceutical Sciences
- Nanotechnology
- Drug Delivery Systems
Background:
- Simvastatin is a widely used statin for hypercholesterolemia.
- Nanoemulsions offer improved drug solubility and bioavailability.
- Developing stable and safe nanoformulations is crucial for effective drug delivery.
Purpose of the Study:
- To prepare and characterize simvastatin-loaded nanoemulsions (SIM-LN).
- To evaluate the physicochemical properties and stability of SIM-LN.
- To assess the toxicity of SIM-LN using in vitro and in vivo models.
Main Methods:
- Nanoemulsions loaded with simvastatin were prepared.
- Physicochemical properties including droplet size and encapsulation efficiency were evaluated over 30 days.
- Toxicity was assessed using Vero cell culture and Caenorhabditis elegans models.
Main Results:
- Prepared SIM-LN exhibited an average droplet size of 139 ± 22 nm.
- High encapsulation rate (>98.4%) was achieved.
- Storage at room temperature was found to be optimal, and no toxicity was observed in preliminary assays.
Conclusions:
- Surfactants effectively optimized nanoemulsion properties without inducing toxicity.
- Simvastatin-loaded nanoemulsions demonstrate favorable characteristics and safety for potential therapeutic use.
