Rous Sarcoma Virus (RSV) and Cancer

5.9K
Rous Sarcoma virus or RSV was discovered by F. Peyton Rous in the year 1911 as a filterable transmissible agent that could cause tumors in chickens. He won a Nobel Prize for this discovery in 1966. His experiments clearly demonstrated that some cancers could be caused by infectious agents and led to the discovery of many more cancer-causing viruses in animals as well as humans.
RSV is a retrovirus that contains two copies of a plus-strand  RNA genome. Its genome consists of four main open...

Summary

Transmembrane protease serine 2 (TMPRSS2) and Angiotensin-converting enzyme-2 (ACE2) are key for SARS-CoV-2 infection. Targeting TMPRSS2 may be a viable therapeutic strategy for COVID-19, despite comparable expression levels between sexes.

Area of Science:

  • Virology
  • Molecular Biology
  • Immunology

Background:

  • Severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) causes the COVID-19 pandemic.
  • Angiotensin-converting enzyme-2 (ACE2) is essential for viral entry.
  • Transmembrane protease serine 2 (TMPRSS2) interacts with ACE2 and facilitates viral entry.

Purpose of the Study:

  • To investigate the expression patterns of ACE2 and TMPRSS2 in various human tissues.
  • To explore the potential role of ACE2 and TMPRSS2 in sex-based differences in COVID-19 severity.
  • To identify potential therapeutic targets for COVID-19.

Main Methods:

  • Analysis of publicly available gene expression datasets.
  • Comparison of ACE2 and TMPRSS2 expression levels in different tissues between males and females.
  • Correlation analysis between viral entry factors and disease severity.

Main Results:

  • ACE2 and TMPRSS2 are expressed in multiple human tissues, including the lungs.
  • Highest ACE2 expression is observed in the testes, and highest TMPRSS2 expression in the prostate.
  • Expression levels of ACE2 and TMPRSS2 were comparable between sexes in most tissues analyzed.
  • Differences in lung expression of these proteins do not fully explain gender disparities in COVID-19 severity.

Conclusions:

  • Targeting TMPRSS2 activity and expression is a rational therapeutic strategy for COVID-19.
  • While ACE2 and TMPRSS2 play crucial roles, their expression levels alone may not account for sex differences in COVID-19 severity.
  • Further research into the complex interplay of viral entry factors and host response is warranted.
