This study identified two distinct osteoarthritis (OA) subtypes using cartilage gene expression. These subtypes show different disease mechanisms and clinical features, paving the way for personalized OA treatments.
Area of Science:
- Cartilage biology
- Molecular genetics
- Osteoarthritis research
Background:
- Osteoarthritis (OA) is a degenerative joint disease with complex pathophysiology.
- Identifying distinct OA subtypes is crucial for developing targeted therapies.
Purpose of the Study:
- To classify osteoarthritis (OA) subtypes using cartilage transcriptomic data.
- To characterize the pathophysiological processes and clinical features of identified OA subtypes.
Main Methods:
- Transcriptomic analysis of 66 primary OA patients' cartilage (preserved and lesioned).
- Unsupervised hierarchical clustering and clinical data integration.
- Protein-protein interaction and pathway enrichment analysis of differentially expressed genes.
Main Results:
- Two robust OA subtypes (cluster A and B) were identified from preserved cartilage.
- Cluster B showed upregulated chemokine pathway genes, but downregulated in lesioned cartilage, indicating complex pathophysiology.
- Cluster B patients exhibited higher joint space narrowing (JSN) and lower osteophyte (OP) scores.
Conclusions:
- Whole-transcriptome profiling revealed two distinct OA subtypes with unique pathophysiological and clinical profiles.
- Further research is needed to confirm these subtypes and integrate clinical data for personalized OA treatment strategies.
