Low blood levels of the thyroid hormones — triiodothyronine (T3) and thyroxine (T4) — signal the hypothalamus to release the thyrotropin-releasing hormone (TRH). TRH then reaches the pituitary gland and stimulates the release of thyroid-stimulating hormone(TSH) into the bloodstream.

Background:

  • Thyroid cancer (TC) is the most common endocrine malignancy.
  • Basigin (BSG) is implicated in tumor invasion, metastasis, and angiogenesis.

Purpose of the Study:

  • To investigate changes in BSG expression in thyroid cancer.
  • To explore the underlying mechanisms of BSG's role in TC.

Main Methods:

  • Analyzed BSG expression using immunohistochemistry (IHC), RNA-sequencing (RNA-seq), and microarrays.
  • Performed integrated analysis of 17 independent datasets (1182 TC cases, 437 controls).
  • Conducted Gene Ontology (GO) and KEGG pathway analyses on BSG co-expressed genes and differentially expressed genes (DEGs).

Main Results:

  • BSG protein levels were significantly higher in TC tissues.
  • Meta-analysis confirmed elevated BSG expression in TC (SMD=0.39, p<0.0001).
  • BSG co-expressed genes were enriched in extracellular matrix (ECM), cell adhesion, and cell-cell interaction pathways; nine hub genes were upregulated in TC.

Conclusions:

  • BSG expression is closely correlated with thyroid cancer progression.
  • BSG may influence TC through modulation of ECM, cell adhesion, and cell-cell interactions.
