Summary

Central nervous system insulin synthesis is localized to a small percentage of neurons. This study confirms local insulin production within the brain, specifically in neural cells, not glial cells.

Background:

  • Previous studies reported insulin mRNA in neural tissue, but cellular origin lacked definitive confirmation.

Purpose of the Study:

  • To clarify the cellular origin of insulin within the CNS.
  • To determine if neurons or glia are responsible for insulin synthesis in the brain.

Main Methods:

  • Immunocytochemistry and in situ hybridization were performed on rabbit brain neuron and glia cultures.
  • Monensin was used to modulate secretory activity and assess its impact on insulin staining.

  • A subset of neurons (3-5%) showed positive immunoreactivity for insulin; glial cells were negative.
