Background:

Purpose of the Study:

  • To clarify the function of the ERK7 gene in T. gondii.
  • To investigate the role of TgERK7 in T. gondii invasion and attachment.

Main Methods:

  • Engineered a complemental strain (pUC/TgERK7) from ΔTgERK7 tachyzoites.
  • Utilized electric transfection with pUC/Tgerk7 plasmid.
  • Compared phenotypic changes (invasion, attachment) with ΔTgERK7 and wild-type GT1 strains.

Main Results:

  • TgERK7 protein was successfully re-expressed in ΔTgERK7 tachyzoites.
  • Eradication of TgERK7 led to significantly lower T. gondii invasion at 1h and 2h post-infection (P < 0.05).
  • No statistical difference in attachment was observed at 15 min (P > 0.05).

Conclusions:

  • TgERK7 protein is a key virulence factor in T. gondii.
  • TgERK7 participates significantly in the invasion stage of T. gondii.
  • Reduced invasion due to TgERK7 absence explains slower intracellular proliferation.
