Objectives:
Porphyromonas gingivalis is one of the etiologic agents of chronic periodontitis. Our previous study showed that the use of minimal media for P. gingivalis allowed to isolate novel inhibitors of P. gingivalis growth. However, growth of P. gingivalis in minimal media was not always reproducible.
Methods:
To explain this phenomenon, we analyzed the growth of seven wild-type ATCC 33277 strains and two wild-type W83 strains in 10 minimal media and three complex media.
Results:
All nine strains grew in LF (Lactalbumin-Ferric chloride), GC (bovine γ-immunoglobulin G-Calcium chloride), and newly developed mC (milk-Casein) minimal media. Therefore, LF, GC, and mC could be used as minimal media for P. gingivalis. In contrast, other six minimal media containing bovine serum albumin (BSA) supported the growth of several less strains; among these, two media also showed lack of reproducibility in growth among ATCC 33277 strains. On the other hand, four ATCC 33277 strains grew similarly in all 13 media, but two W83 and other three ATCC 33277 strains grew differently in at least one medium.
Conclusions:
These results suggest that the lack of reproducibility of P. gingivalis growth on minimal media is caused by the presence of BSA, and by differences among the standard strains of P. gingivalis.
Insights
Reproducibility of Porphyromonas gingivalis growth in minimal media is affected by bovine serum albumin (BSA) and strain variability. Specific media like LF, GC, and mC support consistent growth, aiding future research on periodontitis agents.
Area of Science:
- Microbiology
- Oral Health
- Bacterial Pathogenesis
Background:
- Porphyromonas gingivalis is a key etiological agent in chronic periodontitis.
- Previous studies utilized minimal media for P. gingivalis to discover novel inhibitors.
- Inconsistent P. gingivalis growth in minimal media necessitates further investigation.
Purpose of the Study:
- To elucidate the reasons behind the variable reproducibility of P. gingivalis growth in minimal media.
- To identify reliable minimal media formulations for P. gingivalis culture.
- To assess the impact of different media components and P. gingivalis strains on growth consistency.
Main Methods:
- Growth analysis of nine wild-type P. gingivalis strains (seven ATCC 33277, two W83) across 10 minimal and 3 complex media.
- Evaluation of growth reproducibility across different media formulations and P. gingivalis strains.
Main Results:
- Consistent growth of all nine P. gingivalis strains was observed in Lactalbumin-Ferric chloride (LF), bovine γ-immunoglobulin G-Calcium chloride (GC), and milk-Casein (mC) minimal media.
- Minimal media containing bovine serum albumin (BSA) exhibited variable support for strain growth and inconsistent reproducibility.
- Differences in growth patterns were noted among various ATCC 33277 and W83 strains across tested media.
Conclusions:
- The presence of BSA in minimal media contributes to the lack of reproducible P. gingivalis growth.
- Variations among standard P. gingivalis strains (ATCC 33277 and W83) influence growth consistency.
- LF, GC, and mC are identified as reliable minimal media for reproducible P. gingivalis cultivation, crucial for inhibitor discovery.
