We developed BASE, a software to automate recombinant monoclonal antibody (mAb) library analysis. This tool streamlines immunoglobulin sequence data analysis and plasmid confirmation, reducing manual effort in antibody discovery.
Area of Science:
- Immunology
- Bioinformatics
- Molecular Biology
Background:
- Recombinant monoclonal antibody (mAb) repertoire analysis is crucial for understanding autoimmune diseases.
- Current methods for generating patient-derived mAb libraries are labor-intensive and inefficient.
Purpose of the Study:
- To develop an automated software solution for analyzing single-cell immunoglobulin sequence data.
- To streamline the process of generating recombinant monoclonal antibody libraries.
Main Methods:
- Development of BASE software with two modules: aBASE for immunological annotation and cBASE for plasmid sequence identity confirmation.
- Comparison of automated BASE analysis with manual analysis for accuracy and efficiency.
Main Results:
- BASE provides complete data analysis for single-cell immunoglobulin cloning.
- Automated aBASE analysis showed 100% identity with manual analysis for most outputs, and 96% for immunoglobulin isotype determination.
- cBASE confirmed plasmid sequence identity in 89.8% of cases with high accuracy, comparable to manual results.
Conclusions:
- BASE offers an efficient, user-friendly software for complete immunoglobulin sequence data analysis and tracking.
- cBASE's plasmid sequence identity confirmation provides unique functionality, reducing workflow time.
- BASE is available for local installation or online access.
