Cleavage and Blastulation01:33

Cleavage and Blastulation

49.1K
After a large-single-celled zygote is produced via fertilization, the process of cleavage occurs while zygotes travel through the uterine tube. Cleavage is a mitotic cell division that does not result in growth. With each round of successive cell division, daughter cells get increasingly smaller.

Hedgehog Signaling Pathway02:33

Hedgehog Signaling Pathway

9.3K
The Hedgehog gene (Hh) was first discovered due to its control of the growth of disorganized, hair-like bristles phenotype in Drosophila, much like hedgehog spines. Hh plays a crucial role in the development of organs and the maintenance of homeostasis in both invertebrates and vertebrates. However, while Drosophila has only one Hh protein, mammals have multiple functional Hedgehog proteins - Sonic (Shh), Desert (Dhh), and Indian Hedgehog (Ihh). All of these homologous proteins have adapted to...

Role Of Notch Signalling In Intestinal Stem Cell Renewal01:12

Role Of Notch Signalling In Intestinal Stem Cell Renewal

2.3K
Notch signaling was first discovered in Drosophila melanogaster, where it is involved in cell lineage differentiation. Notch signaling regulates the maintenance and differentiation of intestinal stem cells or ISCs by controlling the expression of atonal homolog 1 or Atoh1. Atoh1 directs cells to differentiate into secretory cells.
Direct cell-to-cell contact is needed for the activation of Notch signaling. The signal is initiated when a notch ligand binds to a receptor on an adjacent cell, also...

Zygotic Development And Stem Cell Formation01:10

Zygotic Development And Stem Cell Formation

6.2K
The development of all multicellular organisms starts with the fusion of haploid cells called sperm and egg to form a diploid zygote. A zygote is a totipotent cell that can develop into a complete organism. The zygote undergoes cell division or cleavage to form an 8-cell mass. Until this stage, the cells are spherical, loosely attached, and remain totipotent. Totipotent cells are capable of developing both the embryonic and the extraembryonic tissues. However, as they continue to divide, they...

Tight Junctions01:29

Tight Junctions

6.5K
Tight junctions are molecular seals between cells that prevent the leaking of fluids, ions, and other small solutes across cavities and compartments in multicellular organisms. They are mainly composed of claudin and occludin transmembrane proteins, and other proteins such as tricellulin and JAM (junctional adhesion molecule). All these proteins are 4-pass transmembrane proteins, except JAM, which is a single-pass transmembrane protein belonging to the immunoglobulin superfamily. The...

Area of Science:

  • Reproductive Biology
  • Developmental Biology
  • Cell Biology

Background:

  • Trophectoderm (TE) barrier function is vital for blastocyst development.
  • Claudin7 (CLDN7) regulates intercellular exchange and cell polarity in epithelial cells.
  • The role of CLDN7 in early porcine embryo development remains unexplored.

Purpose of the Study:

  • To investigate the role of CLDN7 in porcine early embryo development.
  • To determine if CLDN7 influences trophectoderm barrier function in porcine embryos.

Main Methods:

  • CLDN7 expression patterns were analyzed during porcine oocyte and early embryo development.
  • CLDN7 was knocked down using microinjected siRNA in porcine embryos.
  • Blastocyst rates, cell counts, gene expression, and TE permeability were assessed.

Main Results:

  • CLDN7 expression increased from the 8-cell stage onwards.
  • CLDN7 knockdown significantly reduced blastocyst rates and cell numbers (total and TE cells).
  • CLDN7 knockdown disrupted tight junction assembly, paracellular sealing, and fluid accumulation in TE.

Conclusions:

  • CLDN7 is essential for porcine blastocyst development.
  • CLDN7 regulates blastocyst development by modulating trophectoderm barrier function.
  • CLDN7 plays a key role in maintaining TE integrity and promoting fluid transport.
