Area of Science:

  • Veterinary Virology
  • Molecular Diagnostics
  • Epidemiology

Background:

  • Bluetongue virus (BTV) causes significant economic losses in ruminants.
  • BTV's serological diversity complicates vaccine development and disease control.
  • Accurate and rapid BTV serotype identification is crucial for effective management.

Purpose of the Study:

  • To adapt existing bluetongue virus serotyping real-time PCR (RT-qPCR) assays for the Luminex xMAP platform.
  • To develop a cost-effective, high-throughput diagnostic tool for simultaneous detection of multiple BTV serotypes.
  • To evaluate the performance of the novel BTV xMAP assay compared to traditional RT-qPCR.

Main Methods:

  • Utilized existing BTV serotype-specific RT-qPCR assays (BTV-1 to BTV-24).
  • Adapted these assays for multiplex detection using the Luminex xMAP bead-based technology.
  • Tested the assay's specificity and sensitivity using reference strains and BTV-positive samples.

Main Results:

  • The BTV xMAP assay successfully detected all 24 tested BTV serotypes.
  • The BTV xMAP assay demonstrated higher sensitivity (87.55%) compared to serotype-specific RT-qPCR (79.85%).
  • The assay provides specific detection of BTV serotypes 1-24 at a reduced cost.

Conclusions:

  • The developed BTV xMAP assay is a novel and effective diagnostic tool for bluetongue virus.
  • This high-throughput assay facilitates rapid BTV serotype identification, especially for large sample volumes.
  • The assay aids in timely epidemiological assessment, informing control and prevention strategies for bluetongue disease.
