    Background:

    • Uropathogenic bacteria, including Proteus species, can cause urinary tract infections.
    • Hemolysins are virulence factors produced by some bacteria that can lyse red blood cells.
    • Escherichia coli hemolysin (HlyA) is a well-characterized virulence factor.

    Purpose of the Study:

    • To investigate the presence and genetic basis of hemolysin production in uropathogenic Proteus vulgaris, Morganella morganii, and Proteus mirabilis.
    • To compare hemolysin determinants with those of Escherichia coli.

    Main Methods:

    • Southern blotting with an E. coli hly gene-specific DNA probe.
    • Immunoblotting to detect hemolysin-like proteins.
    • Molecular cloning of hemolysin determinants using recombinant plasmids.
    • Restriction endonuclease fragment mapping and transposon-mediated mutagenesis (TnphoA).
    • Minicell polypeptide analysis.

    Main Results:

    • A 110-kilodalton polypeptide similar to E. coli HlyA was expressed in hemolytic P. vulgaris and M. morganii.
    • A novel hemolysin determinant (HpmA) from P. mirabilis was cloned and characterized.
    • HpmA, a 140-kilodalton polypeptide, is common in Proteus species but absent in hemolytic M. morganii, E. coli, Citrobacter freundii, and Serratia marcescens.

    Conclusions:

    • Hemolytic uropathogenic P. vulgaris and M. morganii possess genetic determinants similar to E. coli hemolysin genes.
    • A novel hemolysin determinant, HpmA, is present in Proteus species and contributes to virulence in urinary tract infections.
    • The genetic basis of hemolysin production varies among different uropathogenic bacterial species.
