    Summary

The HAIR-AN syndrome, characterized by hyperandrogenism, insulin resistance, and acanthosis nigricans, shows familial patterns. Weight management and oral contraceptives may improve symptoms in affected individuals.

    Area of Science:

    • Endocrinology
    • Genetics
    • Dermatology

    Background:

    • The HAIR-AN syndrome (hyperandrogenism, insulin resistance, acanthosis nigricans) exhibits a familial predisposition.
    • Understanding its natural history and treatment is crucial for affected families.

    Observation:

    • Two families with all female offspring affected by the HAIR-AN syndrome were studied.
    • Syndrome progression was rapid during peripubertal and early reproductive years, becoming variable later.
    • Rapid weight gain exacerbated insulin resistance and hyperandrogenemia.

    Findings:

    • Weight reduction in one subject with postmenarcheal onset improved dermatologic, hyperandrogenic, and insulin resistance features.
    • Oral contraceptives showed potential in ameliorating dermatologic symptoms.

    Implications:

    • Early recognition and intervention, particularly weight management, are key for managing the HAIR-AN syndrome.
    • Further research into genetic and environmental factors influencing syndrome progression is warranted.
    • Pharmacological and lifestyle interventions can impact clinical outcomes.
