Advancements in molecular biology have revolutionized the identification and characterization of bacteria, with multiple methods leveraging DNA sequencing for enhanced precision. As sequencing technologies improve and costs decline, these approaches are increasingly used in clinical, environmental, and evolutionary studies.Multilocus Sequence Typing (MLST) examines several housekeeping genes, essential chromosomal genes encoding cellular functions, to distinguish strains. Area of Science:

  • Food Microbiology
  • Molecular Biology
  • Pathogen Detection

Background:

  • Staphylococcus aureus (S. aureus) is a significant human pathogen.
  • Food contamination by S. aureus poses a global health risk.
  • Virulence factors are crucial for S. aureus pathogenesis.

Purpose of the Study:

  • To develop a rapid and accurate method for detecting major S. aureus toxin genes.
  • To assess the prevalence of specific virulence genes in S. aureus strains.

Main Methods:

  • Utilized Polymerase Chain Reaction (PCR) technology.
  • Employed a PCR plate for simultaneous detection of multiple toxin genes (sea, seb, sec, see, pvl, tsst).

Main Results:

  • Out of 13 S. aureus strains tested, 12 (92.3%) harbored one or more toxin genes.
  • Achieved one-step detection of key S. aureus toxin factors.

Conclusions:

  • The developed PCR method is effective for rapid, one-step detection of major S. aureus virulence genes.
  • This tool aids in preventing outbreaks linked to S. aureus food contamination.
