mRNA Stability and Gene Expression02:51

mRNA Stability and Gene Expression

6.2K
The structure and stability of mRNA molecules regulates gene expression, as mRNAs are a key step in the pathway from gene to protein. In eukaryotes, the half-life of mRNA varies from a few minutes up to several days. mRNA stability is essential in growth and development. The absence of the proteins regulating its stability, such as tristetraprolin in mice, can cause systemic issues, including bone marrow overgrowth, inflammation, and autoimmunity.

Area of Science:

  • Reproductive Biology
  • Molecular Embryology
  • Epigenetics

Background:

  • Maternal mRNA clearance is essential for early embryonic development.
  • RNA N6-methyladenosine (m6A) modification regulates maternal mRNA clearance.
  • Mechanisms of maternal mRNA clearance in farm animals, particularly goats, are poorly understood.

Purpose of the Study:

  • To identify maternal mRNAs during the maternal-to-zygotic transition (MZT) in goats.
  • To investigate the role of m6A reader YTHDF2 in maternal mRNA clearance and early goat embryogenesis.
  • To elucidate the mechanisms underlying ZGA-dependent maternal mRNA clearance.

Main Methods:

  • Differential gene expression analysis to identify maternal mRNAs in goat embryos.
  • YTHDF2 knockdown using RNA interference in goat embryos.
  • Assessment of embryonic development (blastocyst rate) and maternal mRNA levels in YTHDF2 knockdown embryos.
  • Analysis of deadenylase and decapping enzyme expression.

Main Results:

  • Identified 3362 differentially expressed genes as maternal mRNAs during goat MZT.
  • Found increased expression of m6A reader YTHDF2 during goat zygotic genome activation (ZGA).
  • YTHDF2 knockdown impaired maternal mRNA clearance (both M-decay and Z-decay) and reduced blastocyst rates, linked to decreased deadenylase and decapping enzyme expression.

Conclusions:

  • Maternal mRNA clearance in goats is dependent on ZGA.
  • YTHDF2 plays a critical role in goat early embryogenesis by promoting maternal mRNA clearance, potentially via recruitment of deadenylases and decapping enzymes.
  • This study provides insights into stochastic reprogramming during MZT and improving in vitro goat embryo development.
