Background:

  • Toona sinensis is a traditional medicinal plant with various bioactive compounds.
  • Investigating the chemical constituents of Toona sinensis leaves can lead to the discovery of novel therapeutic agents.

Purpose of the Study:

  • To isolate and characterize terpenoids from the ethyl acetate soluble fraction of Toona sinensis leaf extract.
  • To evaluate the cytotoxic and radical scavenging potential of the isolated compounds.

Main Methods:

  • Column chromatography (silica gel, Sephadex LH-20, MCI, ODS) was used for isolation.
  • Spectroscopic data (NMR, MS) and physicochemical methods were employed for structure elucidation.
  • In vitro cytotoxic activity was assessed using the MTT assay against SMMC-7721, A549, and MCF-7 cell lines.
  • Radical scavenging potential was evaluated using the DPPH assay.

Main Results:

  • Eight terpenoids (1-8) were isolated and identified: 8β-hydroxypimar-15-en-19-oic acid methyl ester (1), cedrodorol B (2), 11β-acetoxyobacunol (3), toonayunnanin D (4), toonaciliatone D (5), toonaciliatone A (6), cedrelone (7), and 11β-hydroxygedunin (8).
  • Compound 1 is a novel pimaradienediterpenoid.
  • Compounds 2-7 were reported from this plant for the first time.
  • Compound 1 demonstrated weak cytotoxic activity (IC50 < 40 μmol·L⁻¹) against tested tumor cell lines.
  • Compound 1 exhibited moderate radical scavenging activity (IC50 = 74.3 μmol·L⁻¹).

Conclusions:

  • The study successfully isolated and identified eight terpenoids from Toona sinensis leaves, including one new compound and seven previously unreported from this source.
  • Compound 1, a new pimaradienediterpenoid, possesses weak cytotoxic and moderate radical scavenging properties.
  • These findings contribute to the understanding of Toona sinensis phytochemistry and suggest potential for further investigation of its therapeutic applications.
