High-Resolution Mass Spectrometry (HRMS)

2.1K
The resolution of a mass spectrometer depends on the efficiency of separating ions with different ion masses. The mass of an atom is approximated to the sum of the masses of protons and neutrons inside, considering the masses of protons and neutrons as equal. However, the masses of the proton (1.6726 × 10−24 g) and neutron (1.6749 × 10−24 g) are not truly equal. There is a minor error in the expression of atomic masses relative to the simplest atom of hydrogen. For...

MALDI-TOF Mass Spectrometry

6.2K
Mass spectrometry is a powerful characterization technique that can identify and separate a wide variety of compounds ranging from chemical to biological entities, based on their mass-to-charge ratio (m/z). The instruments that allow this detection, known as mass spectrometers, have three components: an ion source, a mass analyzer, and a detector. These spectrometers differ based on the nature of their ion source and analyzers.
Matrix-assisted laser desorption ionization (MALDI) is a commonly...

Area of Science:

  • Analytical Chemistry
  • Forensic Science
  • Public Health

Background:

  • Illicit drug use poses global health risks due to acute and chronic consumption impacts.
  • Drug checking facilities are crucial for harm reduction, enabling users to identify unknown substances and adulterants.
  • The rise of new psychoactive substances (NPS) necessitates advanced analytical methods to track evolving drug markets.

Purpose of the Study:

  • To develop a high-throughput analytical strategy for qualitative and quantitative analysis of drug checking samples.
  • To improve the identification of novel and known illicit substances, adulterants, and contaminants.
  • To reduce analysis time compared to conventional methods for faster harm reduction.

Main Methods:

  • Development of a matrix-assisted laser desorption/ionization (MALDI) high-resolution mass spectrometry (HRMS) method.
  • Utilized an online m/z cloud library for untargeted compound identification.
  • Analyzed 67 drug checking samples, comparing results with routine analytical approaches.

Main Results:

  • Successfully identified 10 designer drugs, common drugs of abuse (cocaine, MDMA), adulterants, and contaminants.
  • The MALDI-HRMS strategy demonstrated comparable performance to conventional methods.
  • Achieved a six-fold reduction in analysis and sample preparation time.

Conclusions:

  • MALDI-HRMS is a powerful, high-throughput tool for rapid drug checking analysis.
  • This method can effectively identify emerging substances, supporting timely harm reduction strategies.
  • The accelerated analysis enables quicker responses to changes in the illicit drug market.
