MiR-582-5p inhibits the growth and invasion of osteosarcoma cell by targeting NOVA1

Objective:
To investigate the role of microRNA-582-5p (miR-582-5p) on osteosarcoma (OS) cell behaviors and provide potential therapeutic targets for OS.
Materials And Methods:
Expression levels of miR-582-5p in OS cell lines and normal cell lines were measured by quantitative real-time PCR. Cell proliferation, migration and invasion capacities were assessed by cell counting kit-8 assay, wound-healing assay, and transwell invasion assay, respectively. Downstream target of miR-582-5p was confirmed by Luciferase activity reporter assay and Western blotting assay.
Results:
MiR-582-5p expression was downregulated in OS cell lines compared with normal cell lines. Overexpression of miR-582-5p inhibits OS cell proliferation, migration, and invasion capacities. Neuro-oncological ventral antigen 1 (NOVA1) was chosen as target gene of miR-582-5p by bioinformatics analysis and Luciferase reporter assay. Moreover, overexpression of NOVA1 could impair tumor suppression role of miR-582-5p on OS cell behaviors.
Conclusions:
MiR-582-5p exerts tumor-suppressive role on OS cell behaviors via targeting NOVA1 in vitro, which will help us to understand the mechanisms underlying OS progression.
Insights
MicroRNA-582-5p (miR-582-5p) is downregulated in osteosarcoma (OS) and inhibits cancer cell growth. Targeting NOVA1, miR-582-5p suppresses OS progression, offering potential therapeutic strategies.
Area of Science:
- Molecular Biology
- Oncology
- Biochemistry
Background:
- Osteosarcoma (OS) is a primary bone malignancy with aggressive behavior.
- Understanding the molecular mechanisms driving OS progression is crucial for developing effective therapies.
Purpose of the Study:
- To investigate the role of microRNA-582-5p (miR-582-5p) in osteosarcoma (OS) cell behaviors.
- To identify potential therapeutic targets for OS based on miR-582-5p function.
Main Methods:
- Quantitative real-time PCR to measure miR-582-5p expression.
- Cell counting kit-8, wound-healing, and Transwell invasion assays to assess cell proliferation, migration, and invasion.
- Luciferase activity reporter assay and Western blotting to confirm the downstream target of miR-582-5p.
Main Results:
- miR-582-5p expression was significantly downregulated in OS cell lines compared to normal cell lines.
- Overexpression of miR-582-5p inhibited OS cell proliferation, migration, and invasion.
- Neuro-oncological ventral antigen 1 (NOVA1) was identified as a direct target of miR-582-5p, and its overexpression counteracted the tumor-suppressive effects of miR-582-5p.
Conclusions:
- miR-582-5p acts as a tumor suppressor in osteosarcoma by targeting NOVA1.
- These findings elucidate the in vitro mechanisms of OS progression and suggest miR-582-5p as a potential therapeutic target.

Tumor Progression
Colon cancer is one of the best-documented examples of tumor progression. Early mutation in the APC gene in colon cells causes a small growth on the colon wall called a polyp. With time, this polyp grows into a benign, pre-cancerous tumor. Further...
