Impaired interferon-α expression in plasmacytoid dendritic cells in asthma

Asthmatic patients show a weaker interferon-alpha response in plasmacytoid dendritic cells (pDCs) to rhinovirus (RV) and Toll-like receptor 7 (TLR-7) stimulation. This defective innate immunity may explain increased asthma exacerbations in RV-infected individuals.

- TLR-7 stimulation induced higher IFN-α expression in pDCs from normal subjects compared to asthmatic patients; IL-4 pretreatment reduced this difference.
- RV stimulation led to increased pDC IFN-α production in normal subjects at 48 and 96 hours, but not in asthmatic patients.
- Higher baseline IFN-α in pDCs and increased asthma exacerbations were observed in rhinovirus-positive (RV+) patients.
Conclusions:
- Asthmatic patients exhibit a compromised response to TLR-7 stimulation and reduced sustained IFN-α production upon RV exposure in pDCs.
- These findings suggest a defective innate immune response to RV infection in asthma.
- Further research is needed on the modulatory effects of IL-4 on TLR-7-stimulated pDCs.
