We identified 446 essential genes in the plant-commensal bacterium Pseudomonas protegens Pf-5 using transposon sequencing. This study expands essentiality data for plant-associated bacteria and reveals species-specific gene functions crucial for survival.
Area of Science:
- Microbiology
- Genomics
- Bacterial Genetics
Background:
- Gene essentiality studies are common in bacterial pathogens but scarce for plant-associated bacteria.
- Pseudomonas protegens Pf-5 is a beneficial, biocontrol bacterium with limited genome-wide essentiality data.
- Previous research on Pf-5 primarily focused on secondary metabolism and biocontrol mechanisms.
Purpose of the Study:
- To systematically identify essential genes in Pseudomonas protegens Pf-5 using a genome-wide approach.
- To compare the essential gene set of Pf-5 with those of other Pseudomonas species with different lifestyles.
- To evaluate the accuracy of in silico predictions versus in vitro experimental determination of essential genes.
Main Methods:
- Generation of a dense transposon mutant library for P. protegens Pf-5.
- Application of transposon-directed insertion site sequencing (TraDIS) to identify essential genes.
- Comparative analysis of essential gene sets across four Pseudomonas species (P. protegens, P. simiae, P. syringae, P. aeruginosa).
Main Results:
- 446 essential genes were identified in P. protegens Pf-5, primarily involved in fundamental cellular machinery.
- Genes related to nutrient biosynthesis, stress response, and transport were underrepresented in the essential set.
- Significant overlap in essential genes was observed among the four Pseudomonas species, alongside species-specific essential genes.
Conclusions:
- This study provides a comprehensive essential gene set for P. protegens Pf-5, crucial for understanding plant-associated bacteria.
- Comparative analysis highlights conserved and divergent essential genes across Pseudomonas species, reflecting lifestyle adaptations.
- Expanding essentiality studies across diverse bacterial lifestyles enhances understanding of fundamental biological processes for survival and growth.

Transgenic Plants
The first-ever transgenic plant was a tobacco plant developed in 1983 that showed resistance against the tobacco mosaic virus. Since then, many transgenic plants have been developed and commercialized for improving the agricultural, ornamental, and horticultural value of a crop plant. Transgenic...
