A new visual detection method for Barley yellow mosaic virus (BaYMV) was developed using loop-mediated isothermal amplification (LAMP). This rapid and sensitive technique aids in protecting barley crops and ensuring food security.
Area of Science:
- Plant Pathology
- Molecular Biology
- Biotechnology
Background:
- Barley yellow mosaic disease, caused by Barley yellow mosaic virus (BaYMV), poses a significant threat to barley production in Eurasia.
- Early detection of BaYMV is crucial for effective disease management and safeguarding food security.
Purpose of the Study:
- To develop a simple, rapid, specific, sensitive, and visual detection method for BaYMV.
- To establish a one-step reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay for BaYMV detection.
Main Methods:
- Design of specific oligonucleotide primers for BaYMV coat protein gene amplification.
- Optimization of loop-mediated isothermal amplification (LAMP) conditions.
- Development and validation of a one-step reverse transcription LAMP (RT-LAMP) assay with visual detection using a colorimetric dye.
Main Results:
- The developed LAMP/RT-LAMP method demonstrated high specificity, detecting only BaYMV among eight tested plant viruses.
- The RT-LAMP assay was 10 times more sensitive than RT-PCR.
- Visual detection of positive BaYMV samples was achieved through a distinct color change.
Conclusions:
- The established visual RT-LAMP method is a promising tool for rapid and sensitive BaYMV detection.
- This assay can contribute to effective barley crop protection strategies and enhance food security.
- The method's simplicity and visual readout facilitate its application in various settings for disease surveillance.
