Mouse Models of Cancer Study02:43

Mouse Models of Cancer Study

6.2K
Mice have long served as models for studying human biology and pathology because of their phylogenetic and physiological similarity with humans. They are also easy to maintain and breed in the laboratory, and hence, many inbred strains are now available for research. Studies on mice have contributed immeasurably to our understanding of cancer biology.
The development of transgenic, knockout, and knock-in mice has led to an exponential increase in their use as model organisms in research,...

Area of Science:

  • Oncology
  • Genetics
  • Developmental Biology

Background:

  • Modeling Ewing sarcoma presents challenges due to EWS-FLI1 overexpression inducing apoptosis.
  • Identifying a suitable cell-of-origin is crucial for developing a functional Ewing sarcoma mouse model.

Purpose of the Study:

  • To generate a novel mouse model for Ewing sarcoma.
  • To identify and utilize a cell-of-origin tolerant to EWS-FLI1 expression.

Main Methods:

  • Generation of an EWS-FLI1-expressing mouse model.
  • Selection of embryonic chondrogenic progenitor (eSZ) cells harboring Ewing sarcoma precursors.

Main Results:

  • Successfully generated a mouse model for Ewing sarcoma.
  • Identified eSZ cells as a suitable cell-of-origin for EWS-FLI1 expression.

Conclusions:

  • The developed mouse model provides a platform for studying Ewing sarcoma.
  • Embryonic chondrogenic progenitors are a viable cell type for modeling Ewing sarcoma.
