Factors Affecting Dissolution: Polymorphism, Amorphism and Pseudopolymorphism01:21

Factors Affecting Dissolution: Polymorphism, Amorphism and Pseudopolymorphism

522
Polymorphism refers to the existence of a drug substance in multiple crystalline forms, known as polymorphs. Recently, this term has been expanded to include solvates (forms containing a solvent), amorphous forms (non-crystalline forms), and desolvated solvates (forms from which the solvent has been removed).
Some polymorphic crystals possess lower aqueous solubility than their amorphous counterparts, leading to incomplete absorption. For instance, the oral suspension of Chloramphenicol, which...

X-ray Diffraction of Biological Samples01:10

X-ray Diffraction of Biological Samples

4.4K
X-ray diffraction or XRD is an analytical tool that utilizes X-rays to study ordered structures such as crystalline organic and inorganic samples, polycrystalline materials, proteins, carbohydrates, and drugs.
According to Bragg's law, when X-rays strike the sample positioned on a stage, the rays are  scattered by the electron clouds around the sample atoms. The  X-ray diffraction or scattering is caused by constructive interference of the X-ray waves that reflect off the internal...

Pharmaceutical Alternatives: Polymorphic Form-Related and Particle Size-Related Therapeutic Nonequivalence

76
Changes in polymorphic forms can significantly influence the bioavailability of poorly soluble drugs. Although the FDA defines pharmaceutical equivalence based on having the same active ingredient, dosage form, and route of administration, it does not automatically disqualify products with different polymorphic forms. This means two products with different polymorphs can still be deemed pharmaceutically equivalent. However, polymorphic differences can affect properties like wettability,...

Drug Dissolution: Requirements and Profile Comparison01:14

Drug Dissolution: Requirements and Profile Comparison

125
The acceptance criteria for dissolution profile data are anchored in Q values, representing the percentage of drug dissolved within a specified period. This assessment unfolds in three stages:First Stage: The test passes if all six drug dosage units are equal to or greater than Q plus 5%; otherwise, the sample proceeds to the second stage.Second Stage: The average of twelve units must be equal to or greater than Q, with no unit falling below Q - 15% to pass; if not, it progresses to the final...

X-ray Crystallography02:18

X-ray Crystallography

25.2K
The size of the unit cell and the arrangement of atoms in a crystal may be determined from measurements of the diffraction of X-rays by the crystal, termed X-ray crystallography.
Diffraction
Diffraction is the change in the direction of travel experienced by an electromagnetic wave when it encounters a physical barrier whose dimensions are comparable to those of the wavelength of the light. X-rays are electromagnetic radiation with wavelengths about as long as the distance between neighboring...

Area of Science:

  • Pharmaceutical Science
  • Solid-State Chemistry
  • Analytical Chemistry

Background:

  • Losartan potassium, a widely used antihypertensive, exists in multiple polymorphic forms.
  • Form I is the designated pharmaceutical crystal form, but can convert to Form III.
  • Accurate quantification of these polymorphs is essential for drug quality control.

Purpose of the Study:

  • To develop and validate a quantitative method for determining losartan potassium polymorphs.
  • To establish a reliable analytical technique for distinguishing and measuring Form I and Form III.

Main Methods:

  • Recrystallization was used to obtain pure Form I and Form III losartan potassium.
  • Powder X-ray diffraction (PXRD) was the primary technique for characterization and quantification.
  • Calibration curves were generated using characteristic peak area ratios and weight percentages.

Main Results:

  • A linear calibration curve was established for the range of 1-50% (w/w).
  • The method utilized the ratio of characteristic PXRD peak areas for Form I (11.13° 2θ) and Form III (5.64° 2θ).
  • Excellent precision (0.6-4.9%) and a low limit of quantification (2.02% w/w) were achieved.

Conclusions:

  • The developed PXRD method provides a quantitative analysis of losartan potassium polymorph mixtures (Forms I and III).
  • This technique is suitable for controlling the quality of bulk losartan potassium.
  • It represents a novel, consistent, sensitive, and accurate approach for quantifying Form III in losartan potassium samples.
